Quick answer: HPV 66 may be labelled differently across laboratory panels and carcinogenicity frameworks. The test label, evidence classification and your clinical follow-up are not the same thing.

What does an HPV 66 positive result mean?
Some clinical assays include HPV 66 within a high-risk testing panel, while carcinogenicity frameworks have historically described the evidence for HPV 66 differently from established Group 1 carcinogenic genotypes. These labels answer different questions.
HPV 66 positive but Pap smear normal
In the 2025 extended-genotyping pathway, isolated 56/59/66 positivity in primary HPV screening is generally managed with one-year HPV-based repeat testing, provided higher-priority genotypes or high-grade cytology are not present.
What if it is positive again?
A grouped 56/59/66 channel cannot prove HPV 66 persistence unless the assay reports HPV 66 individually.
How should grouped laboratory results be read?
Do not convert “56/59/66 positive” into “HPV 66 positive” unless the laboratory provides individual genotype resolution.
Is HPV 66 covered by vaccination?
HPV 66 is not directly targeted by the 9-valent vaccine.
When can colposcopy or biopsy become relevant?
Colposcopy and biopsy decisions are based on genotype, cytology or dual-stain result where appropriate, prior screening, previous CIN2/3 treatment and the clinical context. HPV genotype alone is not a biopsy diagnosis.
