Quick answer: HPV 56 is a high-risk genotype often discussed within the 56/59/66 group. In selected primary HPV-screening scenarios, one-year HPV-based repeat testing may be appropriate.

What does an HPV 56 positive result mean?
HPV 56 is a carcinogenic high-risk genotype, but its average long-term precancer risk is lower than several higher-priority genotypes. “Lower” does not mean “no risk.”
HPV 56 positive but Pap smear normal
In the 2025 ASCCP extended-genotyping guidance, when primary HPV screening identifies only the 56/59/66 group and no other carcinogenic genotype, one-year HPV-based repeat testing is often recommended. High-grade cytology or prior CIN2/3 changes that pathway.
What if it is positive again?
If the assay reports only a grouped 56/59/66 channel, repeated group positivity does not prove HPV 56 itself persisted.
How should grouped laboratory results be read?
A 56/59/66 positive channel means at least one type in that assay group was detected if the test uses grouped reporting; it does not mean all three types are present.
Is HPV 56 covered by vaccination?
HPV 56 is not directly targeted by the 9-valent vaccine.
When can colposcopy or biopsy become relevant?
Colposcopy and biopsy decisions are based on genotype, cytology or dual-stain result where appropriate, prior screening, previous CIN2/3 treatment and the clinical context. HPV genotype alone is not a biopsy diagnosis.
